Macrophage CCL22 Secretion Under Hypoxic Conditions Promotes the Metastasis of Triple-Negative Breast Cancer

Objective: To explore the mechanism by which macrophages secrete CCL22 to promote metastasis of triple-negative breast cancer under hypoxic conditions. Methods: 20 ng/mL mass concentration phorbol 12-myristate 13-acetate (PMA) cell culture medium, 4?,6-diamidino-2-phenylindole (DAPI), dimethyl sulfoxide (DMSO), trypsin digestion solution, CCL18 Kit , Interleukin (IL)-10 Kit, CCL17 TRIzolTM Reagent Reverse Transcriptase Polymerase Chain Reaction (RT-PCR) cells MDA-MB-231 and BT-549, as well other reagents were used (TNBC) BT-549 human mononuclear THP-1, analyze observe lungs, secretion CCL22, migration cancer, situation CCR4. Results: Compared with normal tumor-associated macrophage (TAM), TAM further promoted tumor cells. The number metastases in induced TAM, was significantly higher than that normoxic TAM. Hypoxia can stimulate expression CCL22. expressions CCR3, CCR4, CCR5 tissues increased compared tissues, CCR4 showed most significant increase. Conclusion: cultured hypoxia enhanced ability TNBC lungs vivo. condition CCL22; receptor, tissues.
